In the presence of doxycycline (100 µg/ml), there was no effect on the expression of the reporter gene in the presence of tetracycline (0.1 µg/ml) (Figure 1). However, the levels of doxycycline in the presence of tetracycline (1, 1, 1, 1.5, 5, or 10 µg/ml) were significantly reduced when tetracycline was added to the medium. This reduction in the levels of doxycycline was most pronounced when tetracycline was added to the medium containing tetracycline at the concentrations of 0.1 and 1 µg/ml, respectively.

In the presence of doxycycline (100 µg/ml), we observed that the expression of the reporter gene was significantly reduced when tetracycline was added to the medium containing tetracycline (0.1 and 1 µg/ml, respectively) (Figure 1). The expression of the reporter gene in the medium was examined by measuring the optical density at OD600nm at tetracycline-inducible plasmid, which was chosen as the tetracycline-inducible plasmid. The optical density was plotted over time to see the change in the expression of the reporter gene at the time of exposure to the tetracycline-inducible plasmid. In the presence of doxycycline, the expression of the reporter gene was significantly increased when tetracycline was added to the medium containing tetracycline at 0.1 µg/ml (Figure 2).

To examine the effect of doxycycline on the expression of the reporter gene, we performed a double-transformed cell line. This cell line was grown in a 5% CO2 incubator with shaking at 25°C. The growth medium was replaced at approximately 8 hours. The expression of the reporter gene was examined by measuring the optical density at OD600nm at tetracycline-inducible plasmid, which was chosen as the tetracycline-inducible plasmid. In the presence of doxycycline, the expression of the reporter gene was significantly increased when tetracycline was added to the medium containing tetracycline at 0.1 µg/ml (Figure 3).

We also examined the effects of tetracycline on the expression of the reporter gene in the presence of doxycycline (0.1, 1, and 2 µg/ml) at a concentration of 100 µg/ml. This concentration of doxycycline was added to the growth medium containing tetracycline at 0.1 µg/ml. As seen in the growth medium, the expression of the reporter gene was significantly increased when the concentration of doxycycline was added to the growth medium containing tetracycline at 0.1 µg/ml (Figure 4). This response was confirmed by the use of a tetracycline-inducible promoter system.
